| dc.description.abstract | O fator de crescimento transformador beta (TGFB) tem papel essencial no equilíbrio da imunidade protetora, imunopatologia e homeostase, e tem sido explorado no contexto do câncer. Porém, evidências sobre sua relação com o câncer do colo do útero (CCU), quarto tipo de câncer mais frequente em mulheres no mundo, associado à presença do Papilomavírus Humano (HPV), são escassas. O objetivo deste trabalho foi avaliar o papel do polimorfismo rs3087465 do receptor do fator de crescimento transformador beta tipo 2 (TGFBR2) na expressão proteica de TGFBR2 e na progressão das lesões intraepiteliais para o CCU, bem como sua associação com a infecção pelo HPV. Adicionalmente, investigar a associação entre alelos, genótipos e haplótipos das variantes rs1800468, rs1800469, rs1800470 e rs1800471 do gene TGFB1 e a expressão proteica com o desenvolvimento do CCU. A primeira etapa deste estudo incluiu 618 mulheres voluntárias, caracterizadas de acordo com o genótipo rs3087465: 302 (48,87%) eram homozigotas para o alelo G, 244 (39,48%) heterozigotas e 72 (11,65%) homozigotas para o alelo A. O genótipo GG foi associado ao desenvolvimento de lesões intraepiteliais escamosas de alto grau (LIEAG) e CCU (p = 0,037), enquanto o genótipo AA foi associado à proteção contra o câncer cervical em análises de regressão logística ajustadas (OR = 0,376; IC 95% = 0,146–0,965; p = 0,042). Não foram observadas associações entre a expressão tecidual de TGFBR2 e os genótipos analisados. Na segunda etapa, 242 mulheres voluntárias foram selecionadas, divididas em grupo controle (sem lesões, NL), composto por mulheres sem evidência citopatológica de lesões intraepiteliais e sem infecção por HPV (n = 190), e grupo caso (câncer cervical, CC), composto por mulheres com carcinoma cervical (n = 52). A combinação das variantes genéticas investigadas resultou em quatro haplótipos com frequências superiores a 5% na população do estudo: GCTG (49,0%), GTCG (32,5%), ACTG (5,9%) e GCCG (5,3%). A distribuição das frequências dos haplótipos entre os grupos revelou que o haplótipo GCTG, sob modelo de herança recessivo, foi associado ao risco de desenvolvimento de CCU (OR = 2,422; IC 95% = 1,006 - 5,834; p = 0,049). | dc.description.abstractother1 | Transforming growth factor beta (TGFB) plays an essential role in the balance of protective immunity, immunopathology, and homeostasis, and has been explored in the context of cancer. However, evidence regarding its relationship with cervical cancer (CCa), the fourth most frequent cancer among women worldwide and associated with the presence of Human Papillomavirus (HPV), is scarce. The aim of this study was to evaluate the role of the rs3087465 polymorphism in the transforming growth factor beta receptor type 2 (TGFBR2) in TGFBR2 protein expression and in the progression of intraepithelial lesions to cervical cancer, as well as its association with HPV infection. Additionally, to investigate the association between alleles, genotypes, and haplotypes of the rs1800468, rs1800469, rs1800470, and rs1800471 variants of the TGFB1 gene and protein expression with the development of cervical cancer. The first stage of this study included 618 female volunteers, characterized according to the rs3087465 genotype: 302 (48.87%) were homozygous for the G allele, 244 (39.48%) were heterozygous, and 72 (11.65%) were homozygous for the A allele. The GG genotype was associated with the development of high-grade squamous intraepithelial lesions (HSIL) and cervical cancer (p = 0.037), whereas the AA genotype was associated with protection against cervical cancer in adjusted logistic regression analyses (OR = 0.376; 95% CI = 0.146–0.965; p = 0.042). No associations were observed between TGFBR2 tissue expression and the analyzed genotypes. In the second stage, 242 female volunteers were selected and divided into a control group (No Lesions, NL), composed of women with no cytopathological evidence of intraepithelial lesions and no HPV infection (n = 190), and a case group (Cervical Cancer, CCa), composed of women with cervical carcinoma (n = 52). The combination of the investigated genetic variants resulted in four haplotypes with frequencies higher than 5% in the study population: GCTG (49.0%), GTCG (32.5%), ACTG (5.9%), and GCCG (5.3%). The distribution of haplotype frequencies between the groups revealed that the GCTG haplotype, under a recessive inheritance model, was associated with the risk of cervical cancer development (OR = 2.422; 95% CI = 1.006 - 5.834; p = 0.049).
