Seleção de microrganismos inibidores do desenvolvimento de fungos em grãos de milho

dc.description.abstractO milho (Zea mays L.) é um dos principais grãos produzidos no Brasil, utilizado como fonte alimentícia animal e humana. Por outro lado, o milho está susceptível à infecção fúngica principalmente durante o período de armazenamento. Os gêneros Aspergillus e Penicillium são predominantes em grãos armazenados e muitas espécies de ambos os gêneros são potenciais produtoras de micotoxinas. Grãos de milho contaminados com micotoxinas, são impróprios para consumo humano e animal, resultando em grandes perdas econômicas. Uma das formas para minimizar a presença de fungos em grãos de milho é utilizar microrganismos que inibem o desenvolvimento de fungos, como algumas espécies do gênero Bacillus. Com este propósito o objetivo desse estudo foi selecionar a ação antagônica de 9 linhagens de Bacillus, com ação antagônica contra A. flavus/ oryzae UELZm4.106. A seleção do potencial antagônico foi feita inicialmente por Cultura Pareada e Difusão em Ágar. Em ambas as triagens foi utilizado 106 células/ mL de Bacillus e 103 conídios/ mL de A. flavus/ oryzae UELZm4.106. Tanto em Cultura Pareada quanto em Difusão em Ágar, somente 4 linhagens de Bacillus, UELAsF5.1, UELAsF4.66, UELAsF1.483 e UELAsA1.447, apresentaram potencial antagônico significativo, com potencial de inibição de 9,6 % até 35,96 %, respectivamente. As 4 linhagens de Bacillus selecionadas foram avaliadas por associação par a par, onde dos 6 tratamentos, apenas 3 pares (UELAsF5.1 + UELAsF4.66, UELAsF5.1 + UELAsA1.447 e UELAsF4.66 + UELAsA1.447) apresentaram inibição de A. flavus/ oryzae UELZm4.106. Os extratos livres de células, das mesmas 4 linhagens de Bacillus, também foram avaliados, porém, não inibiram A. flavus/ oryzae UELZm4.106. As 4 linhagens de Bacillus previamente selecionadas foram avaliadas em grãos de milho. Para tanto, foi utilizado 20 g de grãos de milho que foram submetidos a tratamentos de infecção artificial A. flavus/oryzae UELZm4.106 e tratamento com grãos naturalmente infectados com fungos. Ambos os tratamentos foram expostos às quatro linhagens de Bacillus separadamente e em seguida avaliado o número de conídios. Os resultados obtidos demonstraram ação antagônica das linhagens de Bacillus spp. contra A. flavus/oryzae e contra fungos diversos nos grãos artificialmente infectados (8,25 %) e naturalmente infectados (81,1 % a 88,89 %). Estes dados sugerem que as 4 linhagens de Bacillus apresentam potencial antagônico e são potenciais candidatas ao desenvolvimento de bioinsumos.
dc.description.abstractother1Corn (Zea mays L.) is one of the main grains produced in Brazil, has high nutritional value, and is used as an animal and human food source. On the other hand, corn is susceptible to fungal infection mainly during its storage period. The genera Aspergillus and Penicillium are predominant in stored grains and many species of both genera are potential mycotoxin producers. Corn grains contaminated with mycotoxins are unfit for human and animal consumption, resulting in large economic losses. One of the ways to minimize the presence of fungi in corn grains is to use microorganisms that inhibit the development of fungi, such as some species of the Bacillus genus. For this purpose, the objective of this study was to select the antagonistic action of 9 strains of Bacillus, against A. flavus/oryzae UELZm4.106. The selection of antagonistic potential was initially done by Paired Culture and Agar Diffusion. In both screenings, 106 cells/ mL of Bacillus spp. and 103 conidia/ mL of A. flavus/ oryzae UELZm4.106 were used. Both in Paired Culture and Agar Diffusion, only 4 Bacillus strains, UELAsF5.1, UELAsF4.66, UELAsF1.483 and UELAsA1.447, showed significant antagonistic potential, with inhibition potential from 9.6% to 35.96 %, respectively. The 4 selected Bacillus strains were evaluated by pairwise association, where of the 6 treatments, only 3 pairs (UELAsF5.1 + UELAsF4.66, UELAsF5.1 + UELAsA1.447 and UELAsF4.66 + UELAsA1.447) showed inhibition against A. flavus/ oryzae UELZm4.106. Cell-free extracts from the same 4 Bacillus strains were also evaluated, however, they did not show inhibition against A. flavus/oryzae UELZm4.106. Regarding the antagonistic potential on fungi present in corn grains, 125 mL Erlenmeyer flasks containing 20 g of corn grains were used. The experiments were conducted with 4 controls: autoclaved corn grains, autoclaved corn grains infected by A. flavus/oryzae UELZm4.106 (106 conidia/ mL), corn grains naturally contaminated with fungi and superficially disinfected; autoclaved corn kernels, artificially infected by A. flavus/oryzae UELZm4.106 (106 conidia/ mL) and superficially disinfected. The treatments contained naturally and artificially contaminated corn kernels immersed in a solution containing 108 cells/ mL of each Bacillus spp. The flasks were incubated at 28 °C for 5 days. Furthermore, two additional systems were evaluated where an aliquot of 5 corn grains was removed from each flask before incubation and deposited in Petri dishes containing filter paper; in the other, an aliquot of 3 corn grains was deposited in Petri dishes containing DG18 medium (without antibiotics). The plates were incubated under the same conditions as the flasks. The results obtained in the vials showed that the antagonistic potential of Bacillus spp. against A. flavus/oryzae was similar to that observed by Paired Culture and Agar Diffusion, UELAsF5.1, UELAsF4.66, UELAsF1.483 and UELAsA1.447 was obtained in descending order, respectively. These data suggest that the 4 Bacillus strains have antagonistic potential and are potential candidates for the development of bioinputs.
