Efeitos da nanopartícula de ferrihidrita sobre o estresse oxidativo promovido por herbicidas em ratos wistar

| dc.description.abstract | O uso excessivo de herbicidas pode levar ao estresse oxidativo e tem efeitos prejudiciais à saúde humana. A ferrihidrita, uma nanopartícula de óxido de ferro, mostra-se promissora contra os efeitos tóxicos dos herbicidas, devido à sua alta relação superfície/volume e excelente capacidade de adsorção. Este estudo avaliou se a ferrihidrita previne o estresse oxidativo induzido por herbicidas, por meio de parâmetros relacionados à defesa antioxidante e ao dano oxidativo. Um total de 64 ratos Wistar adultos com peso entre 260-290 g foram divididos em 8 grupos, com 8 ratos em cada grupo. Durante quatro dias consecutivos, os ratos receberam dose diária de 1000 mg/kg de peso corporal de diferentes substâncias: ferrihidrita (F), glifosato (G), Roundup® (R), atrazina (A), ferrihidrita associado ao glifosato (FG), Roundup® (FR) ou atrazina (FA) via gavagem. O grupo controle (C) recebeu água. Após 24 horas a última administração, os ratos foram eutanasiados por decapitação e o fígado, rim e cérebro foram removidos para análise. Os parâmetros avaliados incluíram glutationa-reduzida (GSH, µg.g prot-1), malondialdeido (MDA, pmol.g prot-1), glutationa-S-transferase (GST, nmol CDNB conjugado.min-1.g prot-1), catalase (CAT, µmol H2O2.min-1.g prot-1), proteína carbonil (PCO, nmol carbonil.g prot-1) e acetilcolinesterase (AChE, nmol DTNB.min-1.g prot-1). A ferrihidrita isolada não causou alterações no cérebro e rins; porém promoveu aumento nas concentrações de MDA no fígado que indica peroxidação lipídica neste tecido. O glifosato levou ao aumento da atividade de GST no fígado, a associação com ferrihidrita diminuiu esta atividade em relação ao glifosato isolado, porém esta associação (FG) apresentou aumento nas concentrações de MDA em relação ao controle e ao glifosato; além disso, não houve resultados consistentes com o esperado nos rins, no grupo FG as concentrações de GSH diminuíram em relação aos grupos controle e glifosato. O Roundup® aumentou concentrações de MDA no fígado e no cérebro, em relação ao controle, e a ferrihidrita associada ao Roundup® evitou estes aumentos; além disso, a atividade da CAT diminuiu com administração do Roundup®, no entanto, houve impedimento desta diminuição quando associado com a ferrihidrita. A atrazina aumentou a concentração de GSH no fígado em relação ao controle e o grupo FA impediu este aumento. Concluindo, a ferrihidrita isolada induziu LPO no fígado, mas não nos rins e cérebro. Quando associada ao glifosato, não mostrou resultados consistentes. Porém, quando associada com o Roundup®, foi capaz de impedir o estresse oxidativo, no fígado e no cérebro; por fim, a nanopartícula em associação com a atrazina também pode indicar resultados consistentes em evitar o estresse oxidativo promovido pelo herbicida. | |
| dc.description.abstractother1 | Excessive use of herbicides can lead to oxidative stress and has harmful effects on human health. Ferrihydrite, an iron oxide nanoparticle, shows promise against the toxic effects of herbicides, due to its high surface/volume ratio and excellent adsorption capacity. This study evaluated whether ferrihydrite prevents oxidative stress induced by herbicides, through parameters related to antioxidant defense and oxidative damage. A total of 64 adult Wistar rats weighing between 260-290 g were divided into 8 groups, with 8 rats in each group. For four consecutive days, the rats received a daily dose of 1000 mg/kg of body weight of different substances: ferrihydrite (F), glyphosate (G), Roundup® (R), atrazine (A), ferrihydrite associated with glyphosate (FG), Roundup® (FR) or atrazine (FA) via gavage. The control group (C) received water. 24 hours after the last administration, the rats were euthanized by decapitation and the liver, kidney and brain were removed for analysis. Parameters evaluated included reduced-glutathione (GSH, µg.g prot-1), malondialdehyde (MDA, pmol.g prot-1), glutathione-S-transferase (GST, nmol CDNB conjugated.min-1.g prot-1 ), catalase (CAT, µmol H2O2.min-1.g prot-1), protein carbonyl (PCO, nmol carbonyl.g prot-1) and acetylcholinesterase (AChE, nmol DTNB.min-1.g prot-1. Isolated ferrihydrite did not cause changes in the brain and kidneys; however, it promoted an increase in MDA concentrations in the liver, which indicates lipid peroxidation in this tissue. Glyphosate led to an increase in GST activity in the liver, the association with ferrihydrite decreased this activity in relation to glyphosate alone, however this association (FG) showed an increase in MDA concentrations in relation to the control and glyphosate; Furthermore, there were no results consistent with expectations in the kidneys; in the FG group, GSH concentrations decreased in relation to the control and glyphosate groups. Roundup® increased MDA concentrations in the liver and brain, in relation to the control, and the ferrihydrite associated with Roundup® prevented these increases; Furthermore, CAT activity decreased with the administration of Roundup®, however, this decrease was prevented when associated with ferrihydrite. Atrazine increased the concentration of GSH in the liver in relation to the control and the FA group prevented this increase. In conclusion, ferrihydrite alone induced LPO in the liver, but not in the kidneys and brain. When associated with glyphosate, it did not show consistent results. However, when associated with Roundup®, it was able to prevent oxidative stress in the liver and brain; Finally, the nanoparticle in association with atrazine can also indicate consistent results in avoiding oxidative stress promoted by the herbicide. | |
