Identificação molecular de Scutavirus chelonidalpha5 em tartaruga-de-pente (Eretmochelys imbricata) de vida livre no Brasil

dc.description.abstractA tartaruga-de-pente (Eretmochelys imbricata) é a única espécie de tartaruga presente na costa brasileira que se encontra criticamente ameaçada de extinção globalmente e em perigo nacionalmente. Apesar desta espécie ter distribuição global em águas tropicais, os estudos relacionados ao seu ciclo de vida e suas patologias são ainda muito escassos. A fibropapilomatose (FP) é a doença de maior impacto em tartarugas-marinhas, principalmente nas populações de tartaruga-verde que é a espécie mais afetada. FP é uma doença epizoótica de ocorrência mundial, que promove o crescimento de tumores benignos em todas as áreas do corpo. A etiologia da FP é considerada multifatorialrelacionada à infecção de tartarugas com Chelonid alphaherpesvirus 5 (ChAHV5), atualmente denominado Scutavirus chelonidalpha5. Também foi relacionada à infecção com Caretta caretta papillomavirus 1 (CcPV-1) e Chelonia mydas papillomavirus 1 (CmPV-1). O objetivo deste estudo foi identificar ChAHV5 em tartarugas-de-pente de vida livre, capturadas intencionalmente no Recife de Coroa Vermelha, Bahia, região nordeste do Brasil. Foram capturados 13 animais que passaram por exames físicos, morfométricos e colheita de sangue, fragmentos de pele e de swabs ocular e cloacal. O DNA extraído do material biológico foi submetido à técnica de PCR para a detecção dos genes UL27 (glicoproteína B) e UL30 (DNA polimerase) de ChAHV5 e E1 de papilomavírus. Em dois indivíduos, um com lesão de FP e outro sem, foi obtida a amplificação parcial do gene que codifica a DNA polimerase de ChAHV5. Análises filogenéticas realizadas na sequência de nucleotídeos dos amplicons revelaram que as cepas de ChAHV5 identificadas neste estudo em E. imbricata diferem das já descritas no Brasil em outras espécies de tartarugas marinhas. Os animais positivos para ChAHV5, apesar de estarem com boa condição corporal, apresentaram-se com hipocalcemia, hipomagnesemia, diminuição nos valores de globulina, sódio e linfócitos. Valores aumentados de eosinófilos, heterófilos, ácido úrico, albumina, além de um aumento acentuado de ferro sérico também foram encontrados. Este é o primeiro relato da identificação de ChAHV5 e de lesão de FP nesta espécie de tartaruga marinha de vida livre na costa brasileira. Os achados sugerem uma dieta deficiente ou incapacidade de assimilar os nutrientes, e um sistema imune comprometido. Foi observado anemia na tartaruga CV478, podendo ser secundária FP e leucopenia na tartaruga CV477, possivelmente secundária a má-nutrição ou a infecção viral.
dc.description.abstractother1The hawksbill turtle (Eretmochelys imbricata) is the only sea turtle species present along the Brazilian coast that is classified as critically endangered globally and endangered nationally. Despite its global distribution in tropical waters, studies related to its life cycle and pathologies remain scarce. Fibropapillomatosis (FP) is the most impactful disease affecting sea turtles, particularly green turtle populations, which are the most affected species. FP is an epizootic disease of global occurrence, characterized by the growth of benign tumors across all areas of the body. The etiology of FP is considered multifactorial. Regarding its infectious etiology, FP has been associated with the infection of turtles by Chelonid alphaherpesvirus-5 (ChAHV5), currently designated as Scutavirus chelonidalpha5 (ChAHV5), and has also been linked to infection with Caretta caretta papillomavirus 1 (CcPV-1) and Chelonia mydas papillomavirus 1 (CmPV-1).This study aimed to identify ChAHV5 in free-ranging hawksbill turtles intentionally captured at Coroa Vermelha Reef, Bahia, in the Northeastern region of Brazil. Following the capture of 13 individuals, physical examinations were performed, along with morphometric data collection and the sampling of blood, skin fragments, and ocular and cloacal swabs. DNA extracted from the biological materials was subjected to PCR for the detection of the UL27 (glycoprotein B) and UL30 (DNA polymerase) genes of ChAHV5, as well as the E1 gene of papillomaviruses. In two individuals, one presenting FP lesions and another asymptomatic, partial amplification of the ChAHV5 DNA polymerase gene was achieved. Phylogenetic analyses of the nucleotide sequences from the amplicons revealed that the ChAHV5 strains identified in this study from E. imbricata differed from those previously described in Brazil in other sea turtle species. Animals positive for ChAHV5, despite being in good body condition, showed low levels of calcium, magnesium, globulin, sodium, and lymphocytes. They also presented high levels of eosinophils, heterophils, uric acid, albumin, and marked increase in serum iron. This is the first report of the identification of ChAHV5 and FP lesions in this free-ranging sea turtle species along the Brazilian coast. This indicates that they may have a poor diet or difficulty absorbing nutrients, as well as a weakened immune system. One turtle (CV478) had anemia, possibly due to FP, and another (CV477) had low white blood cell count, perhaps due to malnutrition or viral infection.
