| dc.description.abstract | A soja [Glycine max (L.) Merr.] é uma oleaginosa de importância econômica mundial, rica em aminoácidos e proteínas, é considerada uma das principais fontes para alimentação humana e animal. Mais de 90% da produção é destinada para produção de farelo de soja, o qual é usado na alimentação animal. Aproximadamente 50% da dieta de suínos e 26% de aves são baseadas no consumo de soja. Entretanto, apesar dos aspectos positivos, a soja possui fatores antinutricionais (FAs) que podem afetar a saúde animal, reduzir a qualidade nutricional do farelo e prejudicar a conversão alimentar. Os inibidores de proteases constituem o fator antinutricional mais importante para a soja. O inibidor de tripsina do tipo Kunitz (KTI) é responsável por inibir o funcionamento da tripsina, considerado o FA mais importante na alimentação de animais monogástricos. A tripsina é uma importante enzima animal, envolvida na digestão de diversas proteínas. A inibição da tripsina pode reduzir o crescimento animal entre 30 a 50% devido à hipertrofia/hiperplasia do pâncreas. O KTI no farelo de soja é desativado na indústria por aquecimento. Esse processamento, no entanto, reduz o valor nutricional do farelo pela destruição térmica dos aminoácidos e aumenta o custo energético da produção em 25%. O uso de variedades com baixo KTI ou livre de KTI pode reduzir custos no processamento e também, resultar em melhoria na conversão alimentar dos animais. Para atingir esse objetivo, neste trabalho realizou-se o silenciamento gênico via CRISPR/Cas (Clustered Regularly Interspaced Short Palindromic Repeats/CRISPR-associated proteins), do gene KTI1 na soja, que é um dos responsáveis pela produção do inibidor de tripsina do tipo Kunitz. Para obtenção das plantas editadas, realizou-se a caracterização do gene alvo por meio de ferramentas da bioinformática, seguido do desenho do RNA guia, o qual foi clonado em vetor contendo os elementos da maquinaria CRISPR/Cas. A transformação genética foi realizada por Agrobacterium tumefaciens. As plantas transformadas tiveram a região alvo da edição sequenciadas, a fim de, identificar a ocorrência de mutações. Como resultado foi possível obter plantas editadas com diferentes frequências de edição. Por meio de avanço de geração espera-se obter uma planta estável e em homozigose. | |

Merr.] is an oilseed of global economic importance, rich in amino acids and proteins, and is considered one of the main sources for human and animal nutrition. More than 90% of production is destined for the production of soybean meal, which is used in animal feed. Approximately 50% of the diet of pigs and 26% of poultry is based on soy consumption. However, despite the positive aspects, soy has anti-nutritional factors (ANF'S) that can affect animal health, reduce the nutritional quality of the bran and impair feed conversion. Protease inhibitors constitute the most important antinutritional factor for soybeans. The kunitz-type trypsin inhibitor (KTI) is responsible for inhibiting the functioning of trypsin, considered the most important ANF in the diet of monogastric animals. Trypsin is an important animal enzyme, involved in the digestion of various proteins. Trypsin inhibition can reduce animal growth by 30 to 50% due to pancreas hypertrophy/hyperplasia. The KTI in soybean meal is deactivated in industry by heating. This processing, however, reduces the nutritional value of the bran through the thermal destruction of amino acids and increases the energy cost of production by 25%. The use of low-KTI or KTI-free varieties can reduce processing costs and also result in improved feed conversion for animals. To achieve this objective, in this work gene silencing was carried out via CRISPR/Cas (Clustered Regularly Interspaced Short Palindromic Repeats/CRISPR-associated proteins), of the KTI1 gene in soybean, which is one of those responsible for the production of trypsin inhibitor type Kunitz. To obtain the edited plants, the target gene was characterized using bioinformatics tools, followed by the design of the guide RNA, which was cloned into a vector containing the elements of the CRISPR/Cas machinery. Genetic transformation was carried out by Agrobacterium tumefaciens. The transformation plants had the editing target region sequenced in order to identify the occurrence of mutations. As a result, it was possible to obtain plants edited with different editing frequencies. Through generation advancement, it is expected to obtain a stable and homozygous plant. | |
