dc.description.abstract | Resumo: O fruto Syzygium cumini conhecido popularmente como Jambolão apresenta propriedades medicinais devido à presença de metabólitos secundários, dentre os quais se destacam os compostos fenólicos, que possuem atividade farmacológica como antioxidante e antimicrobiana O presente trabalho teve como objetivo investigar os compostos químicos do fruto Jambolão, avaliar seu potencial antioxidante, citotóxico e sua atividade antimicrobiana frente a células sésseis e planctônicas Foram obtidos cinco extratos (EBE1, EBE7, EBE5, EBE3, EBA1) da polpa e casca do fruto Os extratos foram submetidos às análises para quantificar compostos fenólicos, flavonoides totais e avaliar sua atividade antioxidante pelos métodos de DPPH, ABTS, FRAP e quelação de íon ferro, visando determinar o melhor solvente que extraísse metabólitos secundários com potencial antioxidante O extrato EBE1 apresentou os melhores resultados nas análises realizadas demonstrando um teor de compostos fenólicos de 1,92 mgGAEg-1; Flavonoides totais de 4,2 mgRTEg-1, e valores de IC5 de 111,99 µgmL-1, 83,55 µgmL-1 e 3,22µgmL-1 para DPPH, ABTS e Batofenantrolina e para FRAP 98,97 µmol/L eq trolox/µgmL-1 Os principais compostos do extrato EBE1 foram identificados por ESI-MS/MS pela técnica de MRM nos modos positivos e negativos, indicando a presença de Ácido gálico, Ácido p-hidroxibenzóico, Malvidina-3-O-glicosídeo, Petunidina-3-O-glicosídeo, Delfinidina-3-O-glicosídeo, Cianidina-3-O-glicosídeo, Malvidina-3,5-O-diglicosídeo e Petunidina-3,5-O-diglicosídeo Os resultados de citotoxicidade para eritrócitos humanos tratados com extrato EBE1 apresentou atividade hemolítica baixa mesmo na mais alta concentração testada de 1 µgmL-1 com 3,97% de hemólise O extrato EBE1 não apresentou atividade antimicrobiana no teste por difusão em ágar e o ensaio de microdiluição seriada determinou uma concentração inibitória mínima (CIM) acima de 2 µgmL-1 frente às cepas de Staphylococcus aureus, Escherichia coli e Klebsiella pneumoniae A avaliação para biofilme de Ecoli em formação mostrou que o extrato EBE1 nas concentrações de 5, 1 e 2 µg/mL apresentaram redução de biomassa em 48,18%, 64,14% e 8,9% e para células viáveis uma redução de 17,51%, 25,8% e 37,59%, respectivamente Para biofilme formado os resultados obtidos nos tratamentos com o extrato EBE1 em 24 e 48 horas foram de 1,56% e 2,43% (25 µg/mL), 2,49% e 2,95% (5 µg/mL), 2,47% e 4,14% (1 µg/mL), 3,83% e 4,19% (2 µg/mL) e de 4,46% e 6,21% (4 µg/mL), respectivamente Portanto, os resultados sugerem que o extrato EBE1 do fruto jambolão apresenta compostos bioativos que podem ser atribuídos a atividade antioxidante e antibiofilme | pt_BR |
dc.description.abstractother1 | Abstract: The fruit Syzygium cumini popularly known as Jambolão presents medicinal properties due to the presence of secondary metabolites Among them, the phenolic compounds stand out because they are responsible for the pharmacological activity as antioxidant and antimicrobial The present work aimed to investigate the chemical compounds of the Jambolão fruit, to evaluate its antioxidant potential, cytotoxichy and its antimicrobial activity against sessile and planktonic cells Five extracts (EBE1, EBE7, EBE5, EBE3, EBA1) were obtained from fruit pulp and peel The extracts were submitted to the assays to quantify phenolic compounds, total flavonoids and to evaluate their antioxidant activity by the methods of DPPH, ABTS, FRAP and iron ion chelation, in order to determine the best solvent that extracts secondary metabolites with antioxidant potential The EBE1 extract showed the best results in the analyzes performed demonstrating a phenolic compound content of 192 mgGAEg-1; Total flavonoids of 42 mgRTEg-1, and IC5 values of 11199 µgmL-1, 8355 µgmL -1, 9897 µmol/L eq Trolox/µgmL-1 and 322 µgmL-1 for DPPH, ABTS, FRAP and Bataphenanthroline, respectively The major compounds of the EBE1 extract were identified by ESI-MS / MS by the MRM technique in positive and negative modes, indicating the presence of Gallic Acid, p-hydroxybenzoic Acid, Malvidine-3-O-glycoside, Petunidin-3-O- Glycoside, Delfinidine-3-O-glycoside, Cyanidin-3-O-glycoside, Malvidine-3,5-O-diglycoside and Petunidine-3,5-O-diglycoside The cytotoxicity results for human erythrocytes treated with EBE1 extract showed low hemolytic activity even at the highest tested concentration of 1 µgmL-1 with 397% hemolysis The EBE1 extract did not show antimicrobial activity in the agar diffusion test and the serial microdilution assay determined a minimum inhibitory concentration (MIC) above 2 µgmL-1 against Staphylococcus aureus, Escherichia coli and Klebsiella pneumoniae strains The evaluation of the Ecoli biofilm in formation showed that the EBE1 extract at the concentrations of 5, 1 and 2 µg/mL had a biomass reduction of 4818%, 6414% and 89% and for viable cells one Reduction of 1751%, 258% and 3759%, respectively For the obtained biofilm, the results in the treatments with the extract EBE1 in 24 and 48 hours were of 156% and 243% (25 µg/mL), 249% and 295% (5 µg/mL), 247% and 414% (1 µg/mL), 383% and 419% (2 µg/mL) and 446% and 621% (4 µg/mL), respectively Therefore, the results suggest that the EBE1 extract of the jambolão fruit presents bioactive compounds that can be attributed to antioxidant and anti-biofilm activity | pt_BR |
